Menzies’ Schiff Reagent

Menzies’ Formulation

Solution

MaterialAmount
Pararosanilin0.2g
Distilled water400mL
Hydrochloric acid, 1N4mL
Sodium thiosulfate96g

Preparation

  1. Dissolve the dye in boiling distilled water, then add the sodium thiosulfate and mix well.
  2. Filter, add the hydrochloric acid, then stir until the solution is crystal clear and slightly pink with a pink or white precipitate (about 1-2 hours).
  3. Refrigerate overnight and store in a tightly stoppered bottle in the dark.
  4. Filter before use.

Notes

  • The solution should be colorless or very pale amber. Brown solutions will often stain poorly. Usually these have been made from a batch of basic fuchsin containing too little pararosanilin.

Safety Note

Prior to handling any chemical, consult the Safety Data Sheet (SDS) for proper handling and safety precautions.
