Hollande’s Fixative
Formula

| Water | 220 | mL |
| Cupric acetate | 25 | g |
| Picric acid, sat. aqu. | 780 | mL |
| Formalin, 100% | 100 | mL |
| Acetic acid, glacial | 10 | mL |
- Dissolve the cupric acetate into the water.
- Add the picric acid and filter.
- Add the rest of the ingredients and mix.
- Fixation time should be determined by trial.
Description
Copper fixatives are not often used in modern histology. They have a reputation for good preservation, as the copper present may act as a mordant for some dyes, including hematoxylin. For that reason, nuclear staining may be enhanced.
