Tissue Sample
5µ paraffin sections of formalin fixed tissue are suitable.
Protocol
- Bring sections to water via xylene and ethanol.
- Rinse with distilled water.
- Place in working toluidine blue for 10 minutes.
- Rinse with distilled water.
- Dehydrate, clear and mount with a resinous medium.
Expected Results
- Helicobacter – dark blue
- Nuclei – dark blue
- Background – light blue
Notes
- Thin sections show the organisms best.
- Adjust the working dilution and staining time for best results.
- Hematology buffer tablets are suitable for making the stock solution.
