Materials

Tissue Sample

5µ paraffin sections of neutral buffered formalin fixed tissue are suitable. Other fixatives are usually satisfactory.

Protocol

  1. Bring sections to water via xylene and ethanol.
  2. Stain with hemalum for 5 minutes.
  3. Blue in running tap water.
  4. Immerse in a Coplin jar of carbol fuchsin for 1 hour at 37°C.
  5. Decolorise with acid ethanol for 2-3 minutes.
  6. Rinse with 95% ethanol.
  7. Rinse twice with absolute ethanol, 1-2 minutes each.
  8. Clear with xylene and mount with a resinous medium.

Expected Results

  • Acid alcohol fast organisms – red
  • Background – blue
