Solutions

Weigert's iron hematoxylin

Acridine orange
Acridine orange	0.1	g
Distilled water	100	mL

Tissue sample
5µ paraffin sections of neutral buffered formalin fixed tissue are suitable.

Method

1. Bring sections to water via xylene and ethanol.
2. Place in Weigert's iron hematoxylin for 5 minutes.
3. Wash well with tap water.
4. Place in acridine orange solution for 2 minutes.
5. Wash in tap water for 30 seconds.
6. Dehydrate, clear and mount in a non fluorescent resinous medium.

Expected results
Using a BG 12 exciter filter, and OG 4 (yellow) and/or OG5 (orange) barrier filter
Fungi fluoresce green-red.
 

Notes

1. This is most suitable as a screening method.
